First off, are they required for gettin' Twilit at all? And second, if they're done outside the Farrow Quest, do you still need to do the amount required?

From what I can recall, no you don't need to participate. I think the item is set on a timer (Harru's research timer), which should activate upon meeting her. The timer lasts about 3 days. Afterward, you can visit her and she should be selling it.
