As far as callbacks go, the game does a pretty direct job of saying “The protagonists of these games have absolutely nothing to do with each other.” The only real references I can think of are the brief mention of a previous champion by Kassyra, and the Fox Jewel item, but that’s not a transformative in this game.

Nothing important; what little you need to know about the original CoC to understand CoC2 is explained by either CoC2's intro or Kassyra, though there are still references to be found. The goblin cultist in the Foothills begins fights with almost the same line that the generic goblin enemy in CoC does ("(It's) Time to get fucked, slut!"), and at least two upcoming characters are spiritual successors to characters that appeared in CoC. On a meta level, Kinu (the most well-developed of the Champion of Hawkethorne's kids) was made partially to spite a portion of CoC's fanbase who wanted the option for the Champion of Ingnam to have sex with one of their more well-developed kids (specifically, their kid with Helia, commonly referred to as Helspawn).

Yeah, I haven't seen anything that would really require knowledge of the original game so far, except as easter eggs. It's possible that may change a bit in the future, if only because Calla presumably ended up in Mareth during the prologue so whatever happens to her on the other side might make more sense with knowledge of the first game. But the way the intro and Kas' dialogue have handled references so far, it's probably never going to be required to play the first to understand this game.
